Coronaviruses nucleocapsid proteins (N-proteins) are low-variable and most highly expressed. N-protein level correlates with viral RNA load. The detection of nucleocapsid antigen in a respiratory swab confirms the diagnosis of COVID-19. Negative test result requires re-testing the patient using NAAT.
